The simple win sets the Amsterdammers up in a strong position ahead of the return match next week. If Ajax win again, they will meet either Irish side Shelbourne FC or Estonian club Nõmme Kalju in the third qualification round for the third-tier European competition.
The Amsterdam goal total included one for 16-year-old debutant Mohamed Abdalla, who became the youngest player to score for a Dutch club in Europe.
There was disappointment for FC Twente, who are hoping to qualify for the group stage of the Europa League. They lost 1-2 at home to Hungarian side Ferencváros and will have to pull out all the stops during the away leg next week to remain in the competition.
If they do lose, Twente will move down to the qualification rounds of the Conference League.
PSV Eindhoven and Feyenoord have qualified directly for the Champions League while NEC is taking part in the qualification competition. AZ, as winners of the KNVB Cup last season, will take part in the Europa League competition.
